Targeting and degrading proteins
The presence or absence of specific proteins, in a certain quantity, at a particular time, in certain tissues, control all essential processes in living organisms. This also includes processes that lead to human diseases if they are not controlled properly. Up to now, in medicine, a specific protein was mainly added as a drug to increase the effectiveness of this protein or an inhibitor was used to decrease the effect of a protein. However, it is equally promising for pharmaceutical drug discovery to directly reduce a protein's concentration. Targeting and degrading proteins in the cell is a relatively new mechanism of action in modern therapies. A target protein is tagged for degradation and degraded by the natural ubiquitin-proteasome system. Compounds such as "Molecular Glues" and "Proteolysis Targeting Chimeras" (PROTACs) initiate this process by binding the respective target protein to an E3 ligase. Protein degradation, permeability studies, "ternary complex" formation, ubiquitination, and degradation phenotype studies, are just some areas that will be presented and discussed in this context at the ELRIG.de Forum 2023.
