PELOAcademy | Stem Cell Culture: Differences, Properties, Prospects, Applications
Presented by Prof. Dr. Gerhard Gstraunthaler, Institut für Physiologie der Medizinischen Universität Innsbruck, emer
Stem cells are unique cell populations that retain the capability of either self-renewal in nearly infinite cell divisions or to differentiate into somatic cell types (asymmetric cell division). In this webinar the challenges in stem cell culture techniques, the types and characteristics of stem cells, and their applications and future perspectives will be discussed.

PELOAcademy: Clinical Potential of MSC-EVs and Translational Challenges
Prof. Dr. rer. nat. Bernd Giebel, Universitätsklinikum Essen (AöR), Institut für Transfusionsmedizin
Human mesenchymal stromal cells (MSCs) are a therapeutically relevant, heterogenous cell entity with immunomodulatory and pro-regenerative potentials. Apparently, MSCs mediate a huge proportion of their therapeutic effects via extracellular vesicles (EVs). Connected to several advantages in using cell-free products for the therapeutic setting, MSC-EVs emerged as promising novel therapeutic agent for various diseases, including graft-versus-host disease (GvHD), ischemic stroke, COVID-19, and sepsis. Learn more about a lentiviral, hTERT-based immortalization strategy, EVs and immortalized MSCs (ciMSCs).

Considerations & Testing: Using Primary Hepatocytes in 2D and 3D Cell Culture
Presented by Dr. Rafal P. Witek, a KOL in the hepatocyte sector, hosted by our partner AnaBios
Review selection criteria and sources of variabilities including donor variabilities, process variabilities, and functional stability that impact the performance of cryopreserved primary hepatocytes when used in In Vitro cell culture. Different criteria for 2D and 3D use will be covered to enable participants to understand how to select the best-performing primary hepatocyte lots for their studies. We cover also best practices when handling primary hepatocytes during storage, after thawing, during plating, and during 3D aggregation to overcome challenges related to data reproducibility and variability.

MSCs and Exosomes Therapeutics: Lessons Learned and a Promising Future
Presented by Kevin Hicok, KCH-Cell-Tx Consulting
and hosted by Rob McCarthy, VitaCyte
Clinical research explored the use of mesenchymal stem cells (MSCs) to improve islet engraftment or suppress the immune response to allo-islets. The webinar will address the questions you may have about MSCs such as: what tissues contain MSCs and how are they isolated? How do they work? What is the status of MSCs as the primary or supplementary cell therapy?

PELOAcademy | Brain organoids: A versatile 3D tool to study human neurological development and disease
Presented by Dr. Insa S. Schroeder, GSI Helmholtzzentrum für Schwerionenforschung Darmstadt, Germany
Brain organoids, derived from human pluripotent stem cells, revolutionize neuroscience. The lecture introduces organoids and covers generation techniques, region-specific specialization, and their role in modeling neurological diseases. Explore their potential for disease research, drug development, ethical considerations, and their ability to mimic human brain development.

EMERGE Episode 15 | Analysis of Quantitative MS-based Proteomics Experiments using MSstats and MSstatsTMT”
Presented by Devon Kohler, Khoury College of Computer Sciences at Northeastern University, USA and ReSyn Biosciences, South Africa
In this webinar we will review the core workflow and methods behind MSstats and its extensions, highlighting critical choices that need to be made in each step of the analysis. We will review how different analysis choices affect the final protein-level conclusions made on the experiment and discuss how to avoid potential analysis pitfalls.

Certificates of Analysis for Tissue Dissociation Enzymes: Practical Utility
Presented Robert McCarthy, VitaCyte, USA
Enzyme suppliers provide Certificates of Analysis (CoAs) for each lot of product purchased by the customer. The primary enzyme manufacturer or the companies re-selling the manufacturer’s product use these CoAs to report the results of quality testing and acceptance criteria used to release the product for sale. The specification limits set internally by the manufacturer are guided by customer feedback or by internal decisions to ensure the majority of lots pass specifications.

PELOAcademy: Why the Best Suitable Media is Important for Exosome Production
Presented by Hyungtaek Jeon, Xcell Therapeutics, Institute of Life Science, Korea
Mesenchymal stem cells (MSCs) are commonly used as a source of regenerative medicine due to their strong immunosuppressive and regenerative effects. The paracrine effect is one of the key mechanisms of MSC. Recently, it has been shown that extracellular vesicles (EVs), which include exosomes and microvesicles, orchestrate the principle mechanisms of action of MSCs after infusion.
Join: the discussion what is the ideal culture environment for the production of exosomes.
Expect: MSC-derived exosome productivity is compared using several media including CDM. Wound healing assay and angiogenesis assay were conducted to evaluate the characteristics of the produced exosomes.
