In general, isolated directly from tissue, primary cells resemble normal physiology and do not exhibit the changes seen in immortalized, genetically modified cell lines. However, successful isolation of healthy primary cells requires extensive training, time, money, and human resources for a typical laboratory.  Even then, protocols and results vary, and cells are susceptible to contamination. In addition, mass-produced general-purpose culture media are not optimized for the unique requirements of individual primary cell types.
